Doppel-Strang Pausen geh&ouml;ren zu den kritischsten DNA-L&auml;sionen im Hinblick auf Zelltod und Erhaltung der genomischen Integrit&auml;t. Schnelle Phosphorylierung der Histon-Variante H2AX bei Ser-139 gammaH2AX zu bilden ist eine fr&uuml;he zellul&auml;re Reaktion auf DNA-Doppel-Strang-Pausen. Visualisierung von diskreten gammaH2AX Herde mit Immunfluoreszenz-basierte Assays hat eine sensible und wirksame Methode zum Nachweis von DSBs die verwickelt werden k&ouml;nnen in verschiedenen Krankheiten einschlie&szlig;lich Krebs, altersbedingten Krankheiten, chronischen Entz&uuml;ndungen und Isch&auml;mie-Reperfusion Verletzung zur Verf&uuml;gung gestellt. In dieser &Uuml;berpr&uuml;fung, die potenzielle Dienstprogramm und die Bedeutung der gammaH2AX als molekulare Marker der Alterung und Krankheit werden analysiert.

Double-strand breaks are one of the most critical DNA lesions with respect to cell-death and preservation of genomic integrity. Rapid phosphorylation of the histone variant H2AX at Ser-139 to form gammaH2AX is an early cellular response to DNA double-strand breaks. Visualization of discrete gammaH2AX foci using immunofluorescence-based assays has provided a sensitive and effective method for detecting DSBs which may be implicated in various pathologies including cancer, age-related diseases, chronic inflammatory diseases and ischemia-reperfusion injury. In this review, the potential utility and significance of gammaH2AX as a molecular marker of aging and disease is analysed.

GammaH2AX as a molecular marker of aging and disease.

Double-strand breaks represent an extremely cytolethal form of DNA damage and thus pose a serious threat to the preservation of genetic and epigenetic information. Though it is well-known that double-strand breaks such as those generated by ionising radiation are among the principal causative factors behind mutations, chromosomal aberrations, genetic instability and carcinogenesis, significantly less is known about the epigenetic consequences of double-strand break formation and repair for carcinogenesis. Double-strand break repair is a highly coordinated process that requires the unravelling of the compacted chromatin structure to facilitate repair machinery access and then restoration of the original undamaged chromatin state. Recent experimental findings have pointed to a potential mechanism for double-strand break-induced epigenetic silencing. This review will discuss some of the key epigenetic regulatory processes involved in double-strand break (DSB) repair and how incomplete or incorrect restoration of chromatin structure can leave a DSB-induced epigenetic memory of damage with potentially pathological repercussions.

Obwohl es bekannt ist, dass doppelte wie die von ionisierender Strahlung zu den wichtigsten urs&auml;chlichen Faktoren hinter Mutationen, Chromosomenaberrationen, genetische Instabilit&auml;t und Karzinogenese sind, deutlich weniger &uuml;ber die epigenetischen folgen Doppel-Strang-Pause-Entstehung und Reparatur f&uuml;r Karzinogenese bekannt. Doppel-Strang-Pause-Reparatur ist eine gut koordinierte Prozess, der erfordert das Entwirren der komprimierte Chromatin-Struktur Reparatur Maschinen Zugang und dann die Wiederherstellung des Originalzustands unbesch&auml;digt Chromatin zu erleichtern. Neuere Experimentelle Befunde haben auf einen m&ouml;glichen Mechanismus f&uuml;r Doppel-Strang Pause-induzierte epigenetische silencing hingewiesen. Radiation therapy is a widely used therapeutic approach for cancer. To improve the efficacy of radiotherapy there is an intense interest in combining this modality with two broad classes of compounds, radiosensitizers and radioprotectors. These either enhance tumour-killing efficacy or mitigate damage to surrounding non-malignant tissue, respectively. Radiation exposure often results in the formation of DNA double-strand breaks, which are marked by the induction of H2AX phosphorylation to generate Î³H2AX. In addition to its essential role in DDR signalling and coordination of double-strand break repair, the ability to visualize and quantitate Î³H2AX foci using immunofluorescence microscopy techniques enables it to be exploited as an indicator of therapeutic efficacy in a range of cell types and tissues. This review will explore the emerging applicability of Î³H2AX as a marker for monitoring the effectiveness of radiation-modifying compounds.

Evaluation of the efficacy of radiation-modifying compounds using Î³H2AX as a molecular marker of DNA double-strand breaks.

Strahlentherapie ist eine weit verbreitete therapeutische Herangehensweise an Krebs. Verbesserung die Wirksamkeit der Strahlentherapie, es ist ein intensives Interesse an dieser Modalit&auml;t mit zwei gro&szlig;e Klassen von Verbindungen, Radiosensitizers und Radioprotectors kombinieren. Diese Verbesserung t&ouml;ten Wirksamkeit oder Sch&auml;den an umgebenden nicht-maligner Gewebe, bzw. zu mildern. Strahlenbelastung f&uuml;hrt oft zur Bildung von Br&uuml;chen des Doppel-Strang der DNA, die durch die Induktion von H2AX Phosphorylierung zum Generieren von &gamma;H2AX gekennzeichnet sind. Zus&auml;tzlich zu den grundlegenden DDR kann Signalisierung und Koordinierung der Doppel-Strang Pause Reparatur, die F&auml;higkeit zu visualisieren und Quantifizierung &gamma;H2AX Herde mit Immunfluoreszenz-Mikroskopie-Techniken als Indikator f&uuml;r die therapeutische Wirksamkeit in einer Reihe von Zelltypen und Gewebe ausgenutzt werden. Diesen Bericht werden die aufstrebende Anwendbarkeit der &gamma;H2AX als Marker f&uuml;r die &Uuml;berwachung der Wirksamkeit der Strahlung &auml;ndern erforschen Verbindungen.

There is an intense interest in the development of radiopharmaceuticals for cancer therapy. In particular, radiopharmaceuticals which involve targeting radionuclides specifically to cancer cells with the use of monoclonal antibodies (radioimmunotherapy) or peptides (targeted radiotherapy) are being widely investigated. For example, the ultra-short range Auger electron-emitting isotopes, which are discussed in this review, are being considered in the context of DNAtargeted radiotherapy. The efficient quantitative evaluation of the levels of damage caused by such potential radiopharmaceuticals is required for assessment of therapeutic efficacy and determination of relevant doses for successful treatment. The DNA double-strand break surrogate marker, Î³H2AX, has emerged as a useful biomonitor of damage and thus effectiveness of treatment, offering a highly specific and sensitive means of assessment. This review will cover the potential applications of Î³H2AX in nuclear medicine, in particular radionuclide therapy.

Utility of Î³H2AX as a Molecular Marker of DNA Double-Strand Breaks in Nuclear Medicine: Applications to Radionuclide Therapy Employing Auger Electron-Emitting Isotopes.

Es ist ein intensives Interesse an der Entwicklung von Radiopharmaka f&uuml;r die Krebstherapie. Insbesondere werden Radiopharmaka die Zielgruppenadressierung Radionuklide speziell auf Krebszellen mit dem Einsatz von monoklonalen Antik&ouml;rpern (Radioimmunotherapy) oder Peptide (gezielte Strahlentherapie) umfassen weithin untersucht. Beispielsweise sind die k&uuml;rzeste Reichweite Auger Elektronen ausgebende Isotope, die in diesen Bericht behandelt wurden, im Rahmen der DNAtargeted Strahlentherapie gepr&uuml;ft. Die effiziente quantitative Auswertung des Gehalts an Sch&auml;den, die durch solche m&ouml;glichen radioaktiven Arzneimitteln ist f&uuml;r die Beurteilung der therapeutischen Wirksamkeit und Bestimmung der jeweiligen Dosen f&uuml;r eine erfolgreiche Behandlung erforderlich. Der DNA Doppel-Strang Pause Surrogat-Marker, &gamma;H2AX, entstanden als eine n&uuml;tzliche Biomonitor Sch&auml;den und somit die Wirksamkeit der Behandlung, bietet hochspezifische und sensitive Mittel zur Bewertung. Diesen Bericht werden die Einsatzm&ouml;glichkeiten von &gamma;H2AX in der Nuklearmedizin, in bestimmten Radionuklid-Therapie behandelt.
