Calibration models for the quantitation of commonly used ecstasy substances have been developed using near infrared spectroscopy (NIR) in diffuse reflectance and in transmission mode by applying seized ecstasy tablets for model building and validation. The samples contained amphetamine, N-methyl-3,4-methylenedioxy-amphetamine (MDMA) and N-ethyl-3,4-methylenedioxy-amphetamine (MDE) in different concentrations. All tablets were analyzed using high performance liquid chromatography (HPLC) with diode array detection as reference method. We evaluated the performance of each NIR measurement method with regard to its ability to predict the content of each tablet with a low root mean square error of prediction (RMSEP). Best calibration models could be generated by using NIR measurement in transmittance mode with wavelength selection and 1/x-transformation of the raw data. The models build in reflectance mode showed higher RMSEPs using as data pretreatment, wavelength selection, 1/x-transformation and a second order Savitzky-Golay derivative with five point smoothing was applied to obtain the best models. To estimate the influence of inhomogeneities in the illegal tablets, a calibration of the destroyed, i.e. triturated samples was build and compared to the corresponding data of the whole tablets. The calibrations using these homogenized tablets showed lower RMSEPs. We can conclude that NIR analysis of ecstasy tablets in transmission mode is more suitable than measurement in diffuse reflectance to obtain quantification models for their active ingredients with regard to low errors of prediction. Inhomogeneities in the samples are equalized when measuring the tablets as powdered samples.

The magnitude of heat and salt transfer between the Indian and Atlantic oceans through 'Agulhas leakage' is considered important for balancing the global thermohaline circulation. Increases or reductions of this leakage lead to strengthening or weakening of the Atlantic meridional overturning and associated variation of North Atlantic Deep Water formation. Here we show that modern Agulhas waters, which migrate into the south Atlantic Ocean in the form of an Agulhas ring, contain a characteristic assemblage of planktic foraminifera. We use this assemblage as a modern analogue to investigate the Agulhas leakage history over the past 550,000 years from a sediment record in the Cape basin. Our reconstruction indicates that Indian-Atlantic water exchange was highly variable: enhanced during present and past interglacials and largely reduced during glacial intervals. Coherent variability of Agulhas leakage with northern summer insolation suggests a teleconnection to the monsoon system. The onset of increased Agulhas leakage during late glacial conditions took place when glacial ice volume was maximal, suggesting a crucial role for Agulhas leakage in glacial terminations, timing of interhemispheric climate change and the resulting resumption of the Atlantic meridional overturning circulation.

Past hydrological changes in Africa have been linked to various climatic processes, depending on region and timescale. Long-term precipitation changes in the regions of northern and southern Africa influenced by the monsoons are thought to have been governed by precessional variations in summer insolation. Conversely, short-term precipitation changes in the northern African tropics have been linked to North Atlantic sea surface temperature anomalies, affecting the northward extension of the Intertropical Convergence Zone and its associated rainbelt. Our knowledge of large-scale hydrological changes in equatorial Africa and their forcing factors is, however, limited. Here we analyse the isotopic composition of terrigenous plant lipids, extracted from a marine sediment core close to the Congo River mouth, in order to reconstruct past central African rainfall variations and compare this record to sea surface temperature changes in the South Atlantic Ocean. We find that central African precipitation during the past 20,000 years was mainly controlled by the difference in sea surface temperatures between the tropics and subtropics of the South Atlantic Ocean, whereas we find no evidence that changes in the position of the Intertropical Convergence Zone had a significant influence on the overall moisture availability in central Africa. We conclude that changes in ocean circulation, and hence sea surface temperature patterns, were important in modulating atmospheric moisture transport onto the central African continent.

Langfristige Ver&auml;nderungen der Niederschl&auml;ge in den Regionen der n&ouml;rdlichen und s&uuml;dlichen Afrika, die durch den Monsun beeinflusst werden gedacht, um die vorr&uuml;ckenden Variationen im Sommer Sonnenbestrahlung geregelt. Umgekehrt wurden kurzfristige &Auml;nderungen der Niederschlag in den n&ouml;rdlichen afrikanischen Tropen Nordatlantik Meer Oberfl&auml;chentemperatur Anomalien, beeinflussen die nordw&auml;rts Erweiterung der innertropischen Konvergenzzone und seine zugeordneten Rainbelt verbunden. A detailed reconstruction of West African monsoon hydrology over the past 155,000 years suggests a close linkage to northern high-latitude climate oscillations. Ba/Ca ratio and oxygen isotope composition of planktonic foraminifera in a marine sediment core from the Gulf of Guinea, in the eastern equatorial Atlantic (EEA), reveal centennial-scale variations of riverine freshwater input that are synchronous with northern high-latitude stadials and interstadials of the penultimate interglacial and the last deglaciation. EEA Mg/Ca-based sea surface temperatures (SSTs) were decoupled from northern high-latitude millennial-scale fluctuation and primarily responded to changes in atmospheric greenhouse gases and low-latitude solar insolation. The onset of enhanced monsoon precipitation lags behind the changes in EEA SSTs by up to 7000 years during glacial-interglacial transitions. This study demonstrates that the stadial-interstadial and deglacial climate instability of the northern high latitudes exerts dominant control on the West African monsoon dynamics through an atmospheric linkage.

Calcitonin-sezernierende pankreatischen endokrinen Tumoren: systematische Analyse einer seltenen Tumor-Einheit.

Familial pancreatic cancer (FPC) is a rare hereditary tumor syndrome. The 10-years experience of the national case collection for familial pancreatic cancer of Germany (FaPaCa) is reported. Since 1999 FaPaCa has collected families with at least two first-degree relatives with confirmed pancreatic cancer (PC), who did not fulfill the criteria of other hereditary tumor syndromes. Histopathological verification of tumor diagnoses, and genetic counseling were prerequisites for enrollment of families in FaPaCa. 94 of 452 evaluated families fulfilled the criteria for partaking in FaPaCa. PC represented the sole tumor entity in 38 (40%) families. In 56 families additional tumor types occurred, including breast cancer (nÂ =Â 28), colon cancer (nÂ =Â 20) and lung cancer (nÂ =Â 11). In 70 (74%) families the pattern of inheritance was consistent with an autosomal dominant trait. Compared to the preceding generation, a younger age of onset was observed in the offspring of PC patients (median: 57 vs. 69Â years), indicating anticipation. Mutation analyses of BRCA2, PALB2, CDKN2a, RNASEL, STK11, NOD2, CHEK2 and PALLD, revealed deleterious causative germline mutations of BRCA2 and PALB2 in 2 of 70 (3%) and 2 of 41 (4.9%) German FPC families, respectively. Prospective PC screening with EUS, MRI and MRCP detected precancerous lesions (IPMN, multifocal PanIN2/3) or carcinoma in 5.5% (4 of 72) to 12.5% (9 of 72) of individuals at risk, depending on histological verification. Appropriate inclusion of families at high risk for PC in registries, such as FaPaCa, provides a unique and excellent tool to gain clinical and genetic knowledge of FPC. Focused research projects can be conducted most efficiently, when data of different FPC registries are combined.

Seit 1999 hat FaPaCa Familien mit mindestens zwei Verwandten ersten Grades mit best&auml;tigten Bauchspeicheldr&uuml;senkrebs (PC), gesammelt, die nicht das Kriterium andere heredit&auml;re Tumor-syndrome. Histopathologische Pr&uuml;fung der Tumor diagnostiziert, und genetische Beratung waren Voraussetzungen f&uuml;r die Registrierung von Familien in FaPaCa. 94 von 452 ausgewerteten Familien erf&uuml;llt die Kriterien f&uuml;r die Teilhabe FaPaCa. PC vertreten die alleinige Tumor-Entit&auml;t in 38 (40 %) Familien. Pancreatic cancer is among the most dismal of human malignancies. The 5-year survival rate is lower than 5%. The identification of precursor lesions would be the main step to improve this fatal outcome. One precursor lesions are called pancreatic intraepithelial neoplasia (PanIN) and are graduated in grade 1 to 3, whereas grade 3 is classified as carcinoma in situ. Currently, no reliable, noninvasive imaging technique (e.g., ultrasound, computed tomography, magnet resonance imaging) exists to verify PanINs.

The frequency of intrathymic parathyroid glands (IPGs) in patients undergoing parathyroidectomy for renal hyperparathyroidism (rHPT) varies considerably between 14.8% and 45.3%. Total parathyroidectomy with autotransplantation and subtotal parathyroidectomy are the most accepted surgical procedures to treat patients with rHPT. However, routine bilateral cervical thymectomy (BCT) is still discussed, although controversially.

Murine embryonic stem (ES) cells have unusually long telomeres, much longer than those in embryonic tissues. Here we address whether hyper-long telomeres are a natural property of pluripotent stem cells, such as those present at the blastocyst inner cell mass (ICM), or whether it is a characteristic acquired by the in vitro expansion of ES cells. We find that ICM cells undergo telomere elongation during the in vitro derivation of ES-cell lines. In vivo analysis shows that the hyper-long telomeres of morula-injected ES cells remain hyper-long at the blastocyst stage and longer than telomeres of the blastocyst ICM. Telomere lengthening during derivation of ES-cell lines is concomitant with a decrease in heterochromatic marks at telomeres. We also found increased levels of the telomere repeat binding factor 1 (TRF1) telomere-capping protein in cultured ICM cells before telomere elongation occurs, coinciding with expression of pluripotency markers. These results suggest that high TRF1 levels are present in pluripotent cells, most likely to ensure proficient capping of the newly synthesized telomeres. These results highlight a previously unnoticed difference between ICM cells at the blastocyst and ES cells, and suggest that abnormally long telomeres in ES cells are likely to result from continuous telomere lengthening of proliferating ICM cells locked at an epigenetic state associated to pluripotency.

Hier wenden wir uns, ob hyper-lange Telomere sind eine nat&uuml;rliche Eigenschaft von pluripotenten Stammzellen zu gewinnen, wie die Anwesenden bei der Blastozyste innere Zelle Masse (ICM), oder ob es sich um ein Merkmal, erworben von der in-vitro Expansion des ES-Zellen. Wir finden, dass ICM Zellen Telomer Dehnung w&auml;hrend der in-vitro-Ableitung des ES-Zelllinien unterzogen werden. In-vivo-Analyse zeigt, dass die hyper-lange Telomere der Morula injiziert ES Zellen bleiben, hyper-lange im Stadium der Blastozyste und l&auml;nger als Telomere der Blastozyste ICM. Telomer-Verl&auml;ngerung w&auml;hrend der Ableitung von ES-Zelllinien ist mit einem R&uuml;ckgang der heterochromatic Marken Telomere gleichzeitige. Wir fanden auch umfassendere der Telomer wiederholen Bindung Faktor 1 (TRF1) Telomer-capping Protein in kultivierten ICM Zellen bevor Telomer-Dehnung, mit Ausdruck der Pluripotenz Marker zusammenf&auml;llt auftritt. Diese Ergebnisse legen nahe, dass hohe TRF1 in pluripotente Zellen, am ehesten gew&auml;hrleisten kompetente Kappung der neugebildete Telomere vorhanden sind. Diese Ergebnisse einen zuvor unbemerkt Unterschied zwischen ICM-Zellen bei der Blastozyste und ES Zellen markieren, und schlagen vor, da&szlig; die ungew&ouml;hnlich lange Telomere in ES-Zellen ergeben sich aus der kontinuierlichen Telomer Verl&auml;ngerung der proliferativen ICM-Zellen, die bei einem epigenetische Zustand, die mit Pluripotenz gesperrt werden.

Renal hyperparathyroidism (rHPT) as a consequence of an abnormal calcium balance is a frequent complication in patients with chronic kidney disease (CKD). However, calcium homeostasis is also regulated by calcitonin. The relevance of elevated calcitonin levels in patients with rHPT is unclear. This report presents a case of a patient with CKD and mild rHPT scheduled for thyroidectomy for a suspected medullary thyroid carcinoma (MTC) within a mononodular goiter. A hemithyroidectomy with resection of both adjacent parathyroid glands and unilateral central lymph node dissection was performed. Histopathology revealed no evidence of MTC. The rHPT, calcitonin and pentagastrin test subsequently normalized and follow-up revealed no evidence for MTC within the remaining right thyroid lobe. Elevated calcitonin levels in patients with CKD may reflect a physiological response to rHPT than rather represent MTC. The thresholds for calcitonin levels need to be better defined in affected patients to determine the optimal extent of surgical resection.

Kalzium-Hom&ouml;ostase wird jedoch auch durch Calcitonin reguliert. Die Relevanz der erh&ouml;hten Calcitonin Niveaus bei Patienten mit rHPT ist unklar. Dieser Bericht zeigt einen Fall eines Patienten mit CKD und milden rHPT f&uuml;r Thyreoidektomie f&uuml;r eine vermutete Schilddr&uuml;senkarzinom, medull&auml;res (MTC) in ein Mononodular Kropf geplant. Ein Hemithyroidectomy mit Resektion der angrenzenden Nebenschilddr&uuml;sen und einseitigen zentralen Lymphknoten Dissektion durchgef&uuml;hrt wurde. Histopathologie ergab keine Hinweise auf MTC. Die rHPT, Calcitonin und Pentagastrin-Test anschlie&szlig;end normalisiert und Follow-up offenbarten keine MTC innerhalb der verbleibenden rechten Schilddr&uuml;se-Lappen. Erh&ouml;hten Calcitonin Niveaus bei Patienten mit CKD m&ouml;glicherweise eine physiologische Reaktion auf rHPT zur&uuml;ckzuf&uuml;hren, als vielmehr MTC darstellen. Die Schwellenwerte f&uuml;r Calcitonin Niveaus m&uuml;ssen bei betroffenen Patienten zu bestimmen, das optimale Ausma&szlig; der chirurgische Resektion besser definiert werden.

Die Calcitonin-Ebenen k&ouml;nnen manchmal irre Nebenschilddr&uuml;se Chirurgen bei Patienten mit chronischer Nierenerkrankung und renalen Hyperparathyreoidismus: Bericht eines Falles.

Activating mutations in the gene encoding Î²-catenin have been identified in the paediatric form of human craniopharyngioma (adamantinomatous craniopharyngioma, ACP), a histologically benign but aggressive pituitary tumour accounting for up to 10% of paediatric intracranial tumours. Recently, we generated an ACP mouse model and revealed that, as in human ACP, nucleocytoplasmic accumulation of Î²-catenin (Î²-cat(nc)) and over-activation of the Wnt/Î²-catenin pathway occurs only in a very small proportion of cells, which form clusters. Here, combining mouse genetics, fluorescence labelling and flow-sorting techniques, we have isolated these cells from tumorigenic mouse pituitaries and shown that the Î²-cat(nc) cells are enriched for colony-forming cells when cultured in stem cell-promoting media, and have longer telomeres, indicating shared properties with normal pituitary progenitors/stem cells (PSCs). Global gene profiling analysis has revealed that these Î²-cat(nc) cells express high levels of secreted mitogenic signals, such as members of the SHH, BMP and FGF family, in addition to several chemokines and their receptors, suggesting an important autocrine/paracrine role of these cells in the pathogenesis of ACP and a reciprocal communication with their environment. Finally, we highlight the clinical relevance of these findings by showing that these pathways are also up-regulated in the Î²-cat(nc) cell clusters identified in human ACP. As well as providing further support to the concept that pituitary stem cells may play an important role in the oncogenesis of human ACP, our data reveal novel disease biomarkers and potential pharmacological targets for the treatment of these devastating childhood tumours.

Identification of novel pathways involved in the pathogenesis of human adamantinomatous craniopharyngioma.

Mutationen im Gen f&uuml;r die &beta;-Catenin aktivieren wurden identifiziert in p&auml;diatrischen Form menschlichen Kraniopharyngeom (adamantinomat&ouml;ser Kraniopharyngeom, ACP), eine histologisch gutartige, aber aggressiver Tumor der Hypophyse Bilanzierung von bis zu 10 % der p&auml;diatrischen intrakraniellen Tumore. Vor kurzem wir generiert eine ACP-Mausmodell und ergaben, dass wie in menschlichen ACP, Nucleocytoplasmic Anh&auml;ufung von &beta;-Catenin (&beta;-cat(nc)) und Over-activation von den Wnt/&beta;-Catenin-Signalweg tritt nur ein sehr kleiner Teil der Zellen, die Cluster bilden. Hier kombinieren Maus, Genetik, Fluoreszenz Kennzeichnung und Fluss-Sortierung-Techniken, wir haben diese Zellen aus tumorigenic Maus Schleimdr&uuml;sen isoliert und gezeigt, dass die &beta;-cat(nc) Zellen angereichert sind, Kolonie-bildenden Zellen kultiviert in Stammzell-F&ouml;rderung von Medien und haben l&auml;ngere Telomere, die angibt, mit normalen Hypophyse Vorfahre/Stammzellen (PSCs) Eigenschaften gemeinsam. Global-gen-profiling-Analyse hat ergeben, dass diese &beta;-cat(nc) Zellen hohe sezernierte mitogenen Signale, z. B. Mitglieder der SHH, BMP und FGF-Familie, neben mehreren Chemokinen und ihre Rezeptoren, schl&auml;gt eine wichtige Autocrine/parakrine Rolle dieser Zellen in der Pathogenese der AKP-Staaten und eine wechselseitige Kommunikation mit ihrer Umwelt Ausdr&uuml;cken. Schlie&szlig;lich beleuchten wir die klinische Relevanz dieser Ergebnisse zeigen, die diese Wege auch oben-geregelt in den &beta;-cat(nc)-Zelle-Clustern im menschlichen ACP identifiziert sind. Sowie weitere Unterst&uuml;tzung f&uuml;r das Konzept, dass die Hypophyse Stammzellen in der Onkogenese menschlichen ACP eine wichtige Rolle spielen kann, zeigen unsere Daten neuartige Krankheit Biomarker und m&ouml;gliche pharmakologische Ziele f&uuml;r die Behandlung von diese verheerenden Kindheit-Tumoren.
