Rho GTPases orchestrate signaling pathways leading to cell migration. Their function depends on GTP loading and isoprenylation by geranylgeranyl pyrophosphate (GGpp). In this study, we show that in human T cells, geranylgeranylation-and not GTP loading-is necessary for RhoA-mediated downstream events. As a result of GGpp depletion with the 3-hydroxy-3-methylglutaryl-CoA reductase inhibitor atorvastatin, RhoA was sequestered from the membrane to the cytosol and, notwithstanding increased GTP loading, the constitutive activation of its substrate Rho-associated coiled-coil protein kinase-1 was blocked. In line with this, T cells expressing increased GTP-RhoA failed to form an intact cytoskeleton and to migrate toward a chemokine gradient. In vivo treatment with atorvastatin in the rodent model of multiple sclerosis markedly decreased the capacity of activated T cells to traffic within the brain, as demonstrated by multiphoton analysis. Thus, tethering of RhoA to the membrane by GGpp is determinant for T cell migration and provides a mechanism for preventing T cell infiltration into inflamed compartments by 3-hydroxy-3-methylglutaryl-CoA reductase inhibitors.

Understanding the immune response in the central nervous system (CNS) is crucial for the development of new therapeutic concepts in chronic neuroinflammation, which differs considerably from other autoimmune diseases. Special immunologic properties of inflammatory processes in the CNS, which is often referred to as an immune privileged site, imply distinct features of CNS autoimmune disease in terms of disease initiation, perpetuation, and therapeutic accessibility. Furthermore, the CNS is a stress-sensitive organ with a low capacity for self-renewal and is highly prone to bystander damage caused by CNS inflammation. This leads to neuronal degeneration that contributes considerably to the phenotype of the disease. In this chapter, we discuss recent findings emphasizing the predominant role of the adaptive immune system in the pathogenesis of chronic neuroinflammation, that is, multiple sclerosis (MS) in patients and experimental autoimmune encephalomyelitis (EAE) in rodents. In addition, we report on efforts to translate these findings into clinical practice with the aim of developing selective treatment regimens.

New insights into adaptive immunity in chronic neuroinflammation.

Spezielle immunologische Eigenschaften von entz&uuml;ndlichen Prozessen im ZNS, die oft als immun privilegierte Site bezeichnet wird, bedeuten die unterschiedlichen Features von CNS-Autoimmun-Krankheit in Bezug auf Krankheit Einleitung, Verewigung und therapeutischen Zug&auml;nglichkeit. Dar&uuml;ber hinaus die CNS ist ein Spannung-empfindlichen Organ mit einer niedrigen Kapazit&auml;t f&uuml;r Selbsterneuerung und ist sehr anf&auml;llig f&uuml;r Zuschauer-Sch&auml;den, die durch CNS Entz&uuml;ndung. Dies f&uuml;hrt zu neuronale Degeneration, die wesentlich zu den Ph&auml;notyp der Krankheit beitr&auml;gt. Neue Einblicke in adaptiven Immunit&auml;t in chronischen Neuroinflammation.

In the course of autoimmune CNS inflammation, inflammatory infiltrates form characteristic perivascular lymphocyte cuffs by mechanisms that are not yet well understood. Here, intravital two-photon imaging of the brain in anesthetized mice, with experimental autoimmune encephalomyelitis, revealed the highly dynamic nature of perivascular immune cells, refuting suggestions that vessel cuffs are the result of limited lymphocyte motility in the CNS. On the contrary, vessel-associated lymphocyte motility is an actively promoted mechanism which can be blocked by CXCR4 antagonism. In vivo interference with CXCR4 in experimental autoimmune encephalomyelitis disrupted dynamic vessel cuffs and resulted in tissue-invasive migration. CXCR4-mediated perivascular lymphocyte movement along CNS vessels was a key feature of CD4(+) T cell subsets in contrast to random motility of CD8(+) T cells, indicating a dominant role of the perivascular area primarily for CD4(+) T cells. Our results visualize dynamic T cell motility in the CNS and demonstrate differential CXCR4-mediated compartmentalization of CD4(+) T-cell motility within the healthy and diseased CNS.

Previous proteomic and transcriptional analyses of multiple sclerosis lesions revealed modulation of the renin-angiotensin and the opposing kallikrein-kinin pathways. Here we identify kinin receptor B1 (Bdkrb1) as a specific modulator of immune cell entry into the central nervous system (CNS). We demonstrate that the Bdkrb1 agonist R838 (Sar-[D-Phe]des-Arg(9)-bradykinin) markedly decreases the clinical symptoms of experimental autoimmune encephalomyelitis (EAE) in SJL mice, whereas the Bdkrb1 antagonist R715 (Ac-Lys-[D-betaNal(7), Ile(8)]des-Arg(9)-bradykinin) resulted in earlier onset and greater severity of the disease. Bdkrb1-deficient (Bdkrb1(-/-)) C57BL/6 mice immunized with a myelin oligodendrocyte glycoprotein fragment, MOG(35-55), showed more severe disease with enhanced CNS-immune cell infiltration. The same held true for mixed bone marrow-chimeric mice reconstituted with Bdkrb1(-/-) T lymphocytes, which showed enhanced T helper type 17 (T(H)17) cell invasion into the CNS. Pharmacological modulation of Bdkrb1 revealed that in vitro migration of human T(H)17 lymphocytes across blood-brain barrier endothelium is regulated by this receptor. Taken together, these results suggest that the kallikrein-kinin system is involved in the regulation of CNS inflammation, limiting encephalitogenic T lymphocyte infiltration into the CNS, and provide evidence that Bdkrb1 could be a new target for the treatment of chronic inflammatory diseases such as multiple sclerosis.

Multiple Sclerosis (MS) is a chronic inflammatory demyelinating disease of the central nervous system, in which the myelin sheath has been considered to be the primary target for many years. However, an increasing number of reports have focused on neurodegenerative aspects of the disease pathogenesis. Recent studies in post-mortem MS biopsies and in the animal model Experimental Autoimmune Encephalomyelitis (EAE) have shown that key features of neurodegeneration, i.e. axonal transection, neuronal cell atrophy and neuronal death already occur in early disease phases. Furthermore, it has become clear that irreversible disability correlates stronger with the neuronal affectation than with demyelination. However the cause of neuronal damage still remains elusive, since both demyelination-dependent and direct immune cell-mediated mechanisms have been suggested so far. Here, we summarize the current concepts and recently identified molecular mechanisms of inflammatory neurodegeneration in autoimmune CNS inflammation and highlight the role of different immune cells in the complex network of interactions leading to neuronal damage.

Multiplesklerose (MS) ist eine chronisch-entz&uuml;ndliche demyelinisierende Erkrankung des zentralen Nervensystems, in denen der Myelinschicht galt das prim&auml;re Ziel f&uuml;r viele Jahre sein. Allerdings konzentrierten sich immer mehr Berichte auf Neurodegenerative Aspekte der Pathogenese der Krankheit. Neuere Studien in Post-Mortem-MS Biopsien und im Tiermodell experimentellen autoimmunen Enzephalomyelitis (EAE) zeigten, dass die Hauptmerkmale der Neurodegeneration, d.h. axonale Transection, neuronale Zelle Atrophie und neuronalen Tod bereits in fr&uuml;hen Phasen der Krankheit auftreten. Dar&uuml;ber hinaus wurde deutlich, dass irreversible Behinderung mit mit Demyelinisierung der neuronalen Marotte als st&auml;rker korreliert. Neuronale Schadensursache bleibt jedoch noch tr&uuml;gerisch, da sowohl Demyelinisierung-abh&auml;ngige als auch direkte immune Zell-vermittelten Mechanismen, so weit vorgeschlagen wurden. Hier fassen wir die aktuellen Konzepte und k&uuml;rzlich identifizierten Molekulare Mechanismen der entz&uuml;ndlichen Neurodegeneration in autoimmune Entz&uuml;ndung der CNS und Hervorhebung der Rolle der verschiedenen Immunzellen in der komplexen Netzwerk von Interaktionen zu neuronale Sch&auml;den f&uuml;hrt.

Neurodegeneration in autoimmune Entz&uuml;ndung der CNS.

Cytotoxic CD8(+) T cells are considered important effector cells contributing to neuronal damage in inflammatory and degenerative CNS disorders. Using time-lapse video microscopy and two-photon imaging in combination with whole-cell patch-clamp recordings, we here show that major histocompatibility class I (MHC I)-restricted neuronal antigen presentation and T cell receptor specificity determine CD8(+) T-cell locomotion and neuronal damage in culture and hippocampal brain slices. Two separate functional consequences result from a direct cell-cell contact between antigen-presenting neurons and antigen-specific CD8(+) T cells. (1) An immediate impairment of electrical signaling in single neurons and neuronal networks occurs as a result of massive shunting of the membrane capacitance after insertion of channel-forming perforin (and probably activation of other transmembrane conductances), which is paralleled by an increase of intracellular Ca(2+) levels (within <10 min). (2) Antigen-dependent neuronal apoptosis may occur independently of perforin and members of the granzyme B cluster (within approximately 1 h), suggesting that extracellular effects can substitute for intracellular delivery of granzymes by perforin. Thus, electrical silencing is an immediate consequence of MHC I-restricted interaction of CD8(+) T cells with neurons. This mechanism is clearly perforin-dependent and precedes, but is not causally linked, to neuronal cell death.

Zytotoxische CD8(+) T-Zellen gelten als wichtige Effektorzellen zur neuronalen Sch&auml;den in entz&uuml;ndliche und degenerative CNS-Erkrankungen beitragen. Mit Time-lapse video Mikroskopie und zwei-Photonen-Darstellung in Verbindung mit ganz-Zell-Patch-Clamp-Aufnahmen, zeigen wir hier, dass major Histocompatibility-Klasse ich (MHC ich)-eingeschr&auml;nkte neuronale Antigen-Pr&auml;sentation und T-Zell-Rezeptor-Spezifit&auml;t zu bestimmen, CD8(+) T-Zell-Fortbewegung und neuronale Sch&auml;den in Kultur und hippocampal Gehirn Scheiben. Zwei separate funktionelle Konsequenzen ergeben sich aus einem direkten Zell-Zell-Kontakt zwischen Antigen-pr&auml;sentierenden Neuronen und Antigen-spezifische CD8(+) T-Zellen. (2) Antigen abh&auml;ngigen neuronalen Apoptose kann auftreten, unabh&auml;ngig von Perforin und Mitglieder des Clusters Granzyme B (max. ca. 1 h), darauf hindeutet, dass extrazellul&auml;re Wirkungen von Perforin intrazellul&auml;re Lieferung des Granzymes ersetzen k&ouml;nnen. So ist die elektrische Unterdr&uuml;ckender unmittelbare Folge der MHC ich eingeschr&auml;nkte Interaktion von CD8(+) T-Zellen mit Neuronen. Dieser Mechanismus ist klar Perforin abh&auml;ngig und vorausgeht, aber ist nicht kausal verkn&uuml;pft, zum neuronalen Zelltod f&uuml;hren.

Chronic inflammation in various organs, such as the brain, implies that different subpopulations of immune cells interact with the cells of the target organ. To monitor this cellular communication both morphologically and functionally, the ability to visualize more than two colors in deep tissue is indispensable. Here, we demonstrate the pronounced power of optical parametric oscillator (OPO)-based two-photon laser scanning microscopy for dynamic intravital imaging in hardly accessible organs of the central nervous and of the immune system, with particular relevance for long-term investigations of pathological mechanisms (e.g., chronic neuroinflammation) necessitating the use of fluorescent proteins. Expanding the wavelength excitation farther to the infrared overcomes the current limitations of standard Titanium:Sapphire laser excitation, leading to 1), simultaneous imaging of fluorophores with largely different excitation and emission spectra (e.g., GFP-derivatives and RFP-derivatives); and 2), higher penetration depths in tissue (up to 80%) at higher resolution and with reduced photobleaching and phototoxicity. This tool opens up new opportunities for deep-tissue imaging and will have a tremendous impact on the choice of protein fluorophores for intravital applications in bioscience and biomedicine, as we demonstrate in this work.

Expanding two-photon intravital microscopy to the infrared by means of optical parametric oscillator.

Neuronal damage in autoimmune neuroinflammation is the correlate for long-term disability in multiple sclerosis (MS) patients. Here, we investigated the role of immune cells in neuronal damage processes in animal models of MS by monitoring experimental autoimmune encephalomyelitis (EAE) by using two-photon microscopy of living anaesthetized mice. In the brainstem, we detected sustained interaction between immune and neuronal cells, particularly during disease peak. Direct interaction of myelin oligodendrocyte glycoprotein (MOG)-specific Th17 and neuronal cells in demyelinating lesions was associated with extensive axonal damage. By combining confocal, electron, and intravital microscopy, we showed that these contacts remarkably resembled immune synapses or kinapses, albeit with the absence of potential TÂ cell receptor engagement. Th17 cells induced severe, localized, and partially reversible fluctuation in neuronal intracellular Ca(2+) concentration as an early sign of neuronal damage. These results highlight the central role of the Th17 cell effector phenotype for neuronal dysfunction in chronic neuroinflammation.

Neuronale Sch&auml;den in autoimmune Neuroinflammation ist das Korrelat f&uuml;r langfristige Behinderung bei Patienten mit multipler Sklerose (MS). Hier, untersucht wir die Rolle von Immunzellen in neuronale Sch&auml;den Prozesse in Tiermodellen f&uuml;r MS durch die &Uuml;berwachung der experimentellen autoimmunen Enzephalomyelitis (EAE) mithilfe von zwei-Photonen-Mikroskopie der bet&auml;ubt lebenden M&auml;usen. Im Hirnstamm entdeckt wir kontinuierliche Interaktion zwischen Immun- und neuronale Zellen, besonders w&auml;hrend Krankheit Spitze. Direkte Interaktion von Myelin Oligodendrocyte-Glykoprotein (MOG)-spezifischen Th17 und neuronalen Zellen in demyelinisierende L&auml;sionen wurde mit umfangreichen axonale Sch&auml;den verbunden. Two-photon laser scanning microscopy (TPLSM) has become a powerful tool in the visualization of immune cell dynamics and cellular communication within the complex biological networks of the inflamed central nervous system (CNS). Whereas many previous studies mainly focused on the role of effector or effector memory T cells, the role of naÃ¯ve T cells as possible key players in immune regulation directly in the CNS is still highly debated.

In vivo imaging of lymphocytes in the CNS reveals different behaviour of naÃ¯ve T cells in health and autoimmunity.

Visualizing rodent cerebral vasculature is an important tool in experimental stroke research. Intravascular perfusion with colored latex has been the method of choice until recently. However, latex perfusion has some technical limitations which compromise its reproducibility. We therefore describe a simple and reproducible method to visualize cerebral vessels in mice. A mixture of two commercially available carbon black inks is injected into the thoracic aorta resulting in efficient filling and high contrast visualization of cerebral vessels. Feasibility of this technique has been validated by identifying anastomotic points between anterior and middle cerebral arteries. Furthermore, perfusion with combined carbon inks allows visualization of significantly smaller vessel diameters at a higher vessel density in comparison to perfusion with diluted/undiluted latex. Thus, perfusion with combined carbon inks offers a simple, cost-effective and reproducible technique in order to visualize cerebral vasculature.

Visualization of macroscopic cerebral vessel anatomy--a new and reliable technique in mice.

Nagetier zerebraler Gef&auml;&szlig;e zu visualisieren ist ein wichtiges Instrument in der experimentellen Schlaganfall-Forschung. Intravaskul&auml;re Perfusion mit farbigen Latex ist die Methode der Wahl, bis vor kurzem gewesen. Latex Perfusion hat jedoch einige technischen Beschr&auml;nkungen, die die Reproduzierbarkeit zu gef&auml;hrden. Wir beschreiben daher eine einfache und reproduzierbare Methode um Hirngef&auml;&szlig;e bei M&auml;usen zu visualisieren. Eine Mischung aus zwei handels&uuml;blichen Carbon Black Tinten wird in der thorakalen Aorta, was effiziente Bef&uuml;llung und kontrastreiche Darstellung der Hirngef&auml;&szlig;e injiziert. Durchf&uuml;hrbarkeit dieser Technik ist durch die Identifizierung anastomotic Punkte zwischen vorderen und mittleren Hirnarterien validiert worden. Perfusion mit kombinierten CO2-Farben erlaubt dar&uuml;ber hinaus die Visualisierung der deutlich kleineren Schiffes Durchmesser an einer h&ouml;heren Schiff-Dichte im Vergleich zur Perfusion mit Latex verw&auml;ssert/unverw&auml;ssert. Perfusion mit kombinierten CO2-Tinten bietet somit eine einfache, kosteng&uuml;nstige und reproduzierbare Technik um zerebrale Gef&auml;&szlig;e sichtbar zu machen.

Visualisierung der makroskopischen zerebrale Schiff Anatomie--eine neue und zuverl&auml;ssige Technik in den M&auml;usen.

Vascular endothelial growth factor (VEGF) has potent angiogenic and neuroprotective effects in the ischemic brain. Its effect on axonal plasticity and neurological recovery in the post-acute stroke phase was unknown. Using behavioral tests combined with anterograde tract tracing studies and with immunohistochemical and molecular biological experiments, we examined effects of a delayed i.c.v. delivery of recombinant human VEGF(165), starting 3 days after stroke, on functional neurological recovery, corticorubral plasticity and inflammatory brain responses in mice submitted to 30 min of middle cerebral artery occlusion. We herein show that the slowly progressive functional improvements of motor grip strength and coordination, which are induced by VEGF, are accompanied by enhanced sprouting of contralesional corticorubral fibres that branched off the pyramidal tract in order to cross the midline and innervate the ipsilesional parvocellular red nucleus. Infiltrates of CD45+ leukocytes were noticed in the ischemic striatum of vehicle-treated mice that closely corresponded to areas exhibiting Iba-1+ activated microglia. VEGF attenuated the CD45+ leukocyte infiltrates at 14 but not 30 days post ischemia and diminished the microglial activation. Notably, the VEGF-induced anti-inflammatory effect of VEGF was associated with a downregulation of a broad set of inflammatory cytokines and chemokines in both brain hemispheres. These data suggest a link between VEGF's immunosuppressive and plasticity-promoting actions that may be important for successful brain remodeling. Accordingly, growth factors with anti-inflammatory action may be promising therapeutics in the post-acute stroke phase.

Intracerebroventricularly delivered VEGF promotes contralesional corticorubral plasticity after focal cerebral ischemia via mechanisms involving anti-inflammatory actions.

Vaskul&auml;ren endothelialen Wachstumsfaktor (VEGF) hat potente Angiogenese und neuroprotektive Effekte im isch&auml;mischen Gehirn. Seine Wirkung auf axonale Plastizit&auml;t und neurologische Erholung in der post-acute Strich-Phase war unbekannt. Per Verhaltens-Test kombiniert mit Anterograde Trakt Ablaufverfolgung Studien und immunhistochemische und Molekulare biologische Experimente, untersuchten wir Wirkung einer verz&ouml;gerten i.c.v. Lieferung von rekombinanten menschlichen VEGF(165), ab 3 Tage nach Schlaganfall, funktionellen neurologischen Erholung, Corticorubral Plastizit&auml;t und entz&uuml;ndliche Gehirn Reaktionen bei M&auml;usen, 30 min der mittleren Hirnarterie Okklusion vorgelegt. Wir zeigen hier, dass die langsam progrediente funktionalen Verbesserungen der motor Greifkraft und Koordination, die von VEGF verursacht sind, mit verst&auml;rkten sprie&szlig;en von Contralesional Corticorubral Fasern, die die Pyramidenbahn um &uuml;berqueren der Mittellinie und anregen des Ipsilesional Parvocellular roten Kerns zweigte versehen sind. Infiltrate von CD45 + Leukozyten waren im isch&auml;mischen Striatum von Fahrzeug-behandelten M&auml;usen, die eng Gebiete ausstellen Iba entsprach bemerkt-1 + aktivierte Mikroglia. VEGF abgeschw&auml;cht CD45 + Leukozyten infiltriert um 14 Uhr aber nicht 30 Tage nach Isch&auml;mie und vermindert die Microglial Aktivierung. Insbesondere wurde die VEGF-induzierte entz&uuml;ndungshemmende Wirkung von VEGF ein Medikament aus einer breiten Reihe von inflammatorische Zytokine und Chemokine in beiden Gehirnh&auml;lften zugeordnet. Diese Daten legen nahe, einen Zusammenhang zwischen VEGFs immunsuppressiven und Plastizit&auml;t-F&ouml;rderung der Aktionen, die m&ouml;glicherweise wichtig f&uuml;r erfolgreiche Gehirn Umbau. Dementsprechend m&ouml;glicherweise Wachstumsfaktoren mit Anti-inflammatory Action vielversprechenden Therapeutika in der post-acute Strich-Phase.

Novel therapeutic concepts against cerebral ischemia focus on cell-based therapies in order to overcome some of the side effects of thrombolytic therapy. However, cell-based therapies are hampered because of restricted understanding regarding optimal cell transplantation routes and due to low survival rates of grafted cells. We therefore transplanted adult green fluorescence protein positive neural precursor cells (NPCs) either intravenously (systemic) or intrastriatally (intracerebrally) 6 hours after stroke in mice. To enhance survival of NPCs, cells were in vitro protein-transduced with TAT-heat shock protein 70 (Hsp70) before transplantation followed by a systematic analysis of brain injury and underlying mechanisms depending on cell delivery routes. Transduction of NPCs with TAT-Hsp70 resulted in increased intracerebral numbers of grafted NPCs after intracerebral but not after systemic transplantation. Whereas systemic delivery of either native or transduced NPCs yielded sustained neuroprotection and induced neurological recovery, only TAT-Hsp70-transduced NPCs prevented secondary neuronal degeneration after intracerebral delivery that was associated with enhanced functional outcome. Furthermore, intracerebral transplantation of TAT-Hsp70-transduced NPCs enhanced postischemic neurogenesis and induced sustained high levels of brain-derived neurotrophic factor, glial cell line-derived neurotrophic factor, and vascular endothelial growth factor in vivo. Neuroprotection after intracerebral cell delivery correlated with the amount of surviving NPCs. On the contrary, systemic delivery of NPCs mediated acute neuroprotection via stabilization of the blood-brain-barrier, concomitant with reduced activation of matrix metalloprotease 9 and decreased formation of reactive oxygen species. Our findings imply two different mechanisms of action of intracerebrally and systemically transplanted NPCs, indicating that systemic NPC delivery might be more feasible for translational stroke concepts, lacking a need of in vitro manipulation of NPCs to induce long-term neuroprotection.

Transduction of neural precursor cells with TAT-heat shock protein 70 chaperone: therapeutic potential against ischemic stroke after intrastriatal and systemic transplantation.

Neue therapeutische Konzepte gegen zerebrale Isch&auml;mie-Schwerpunkt zellbasierte Therapien um einige der Nebenwirkungen von Thrombolytische Therapie zu &uuml;berwinden. Zellbasierte Therapien sind jedoch aufgrund der behindert eingeschr&auml;nkt, was die optimale Zelle Transplantation Routen und aufgrund der niedrigen &Uuml;berlebensraten der transplantierten Zellen zu verstehen. Wir daher transplantiert Erwachsene gr&uuml;ne Fluoreszenz Protein positiven neuronalen Vorl&auml;uferzellen (NPCs) entweder intraven&ouml;s (systemischen) oder Intrastriatally (intracerebrally) 6 Stunden nach Schlaganfall bei M&auml;usen. Um das &Uuml;berleben der NPCs zu erh&ouml;hen, wurden Zellen in-vitro-Protein-transduzierten mit TAT-Hitze-Schock-Protein 70 (Hsp70) vor der Transplantation, gefolgt von einer systematischen Analyse der Hirnverletzung und zugrunde liegenden Mechanismen je nach Zelle Zustellrouten. Transduktion von NPCs mit TAT-Hsp70 f&uuml;hrte zu erh&ouml;hten intrazerebrale Zahl der Aufgepfropfte NPCs nach intrazerebralen aber nicht nach systemischen Transplantation. W&auml;hrend systemische Lieferung von systemeigenem oder transduzierten NPCs nachhaltig Neuroprotektion ergab und neurologische Erholung induzierte, verhindert nur TAT-Hsp70-transduzierten NPCs sekund&auml;re neuronale Degeneration nach intrazerebralen Lieferung, die verbesserte funktionelle Ergebnis zugeordnet war. Dar&uuml;ber hinaus intrazerebrale Transplantation von TAT-Hsp70-transduzierten NPCs verbessert postischemic Neurogenese und anhaltend hohe Belastung des Brain-derived Neurotrophic Factor Gliazelle Linie-derived Neurotrophic Factor und vaskul&auml;ren endothelialen Wachstumsfaktor in-vivo induziert. Neuroprotektion nach intrazerebralen Zelle Lieferung korreliert mit der Menge NPCs zu &uuml;berleben. Im Gegenteil, vermittelte systemische Lieferung von NPCs akute Nervenschutz durch Stabilisierung der die Blut-Hirn-Schranke, gleichzeitige mit reduzierten Aktivierung der Matrix Metalloproteinase-9 und verminderte Bildung von reaktiven Sauerstoffspezies. Unsere Ergebnisse bedeuten zwei verschiedene Wirkungsmechanismen von intracerebrally und systemisch transplantierten NPCs, die darauf hinweist, dass systemische NPC Lieferung m&ouml;glicherweise praktikabler f&uuml;r translational Strich Konzepte, fehlt die Notwendigkeit von in-vitro-Manipulation der NPCs, langfristige Nervenschutz induzieren.

Transduktion von neuronalen Vorl&auml;ufer Zellen mit TAT-Hitze-Schock-Protein 70 Anstandsdame: therapeutische Potenzial gegen isch&auml;mischen Schlaganfalls nach Intrastriatal und systemische Transplantation.

Only a minority of stroke patients receive thrombolytic therapy. Therefore, new therapeutic strategies focusing on neuroprotection are under review, among which, inhibition of the proteasome is attractive, as it affects multiple cellular pathways. As proteasome inhibitors like bortezomib have severe side effects, we applied the novel proteasome inhibitor BSc2118, which is putatively better tolerated, and analysed its therapeutic potential in a mouse model of cerebral ischaemia. Stroke was induced in male C57BL/6 mice using the intraluminal middle cerebral artery occlusion model. BSc2118 was intrastriatally injected 12 h post-stroke in mice that had received normal saline or recombinant tissue-plasminogen activator injections during early reperfusion. Brain injury, behavioural tests, western blotting, MMP9 zymography and analysis of angioneurogenesis were performed for up to 3 months post-stroke. Single injections of BSc2118 induced long-term neuroprotection, reduced functional impairment, stabilized blood-brain barrier through decreased MMP9 activity and enhanced angioneurogenesis when given no later than 12 h post-stroke. On the contrary, recombinant tissue-plasminogen activator enhanced brain injury, which was reversed by BSc2118. Protein expression of the transcription factor HIF1A was significantly increased in saline-treated and recombinant tissue-plasminogen activator-treated mice after BSc2118 application. In contrast, knock-down of HIF1A using small interfering RNA constructs or application of the HIF1A inhibitor YC1 (now known as RNA-binding motif, single-stranded-interacting protein 1 (RBMS1)) reversed BSc2118-induced neuroprotection. Noteworthy, loss of neuroprotection after combined treatment with BSc2118 and YC1 in recombinant tissue-plasminogen activator-treated animals was in the same order as in saline-treated mice, i.e. reduction of recombinant tissue-plasminogen activator toxicity through BSc2118 did not solely depend on HIF1A. Thus, the proteasome inhibitor BSc2118 is a promising new candidate for stroke therapy, which may in addition alleviate recombinant tissue-plasminogen activator-induced brain toxicity.

Daher sind neue therapeutische Strategien, die Konzentration auf Neuroprotection im Berichtsjahr, unter denen Hemmung der das Proteasom ist attraktiv, da sie mehrere zellul&auml;ren Wege betrifft. Proteasom-Inhibitoren wie Bortezomib schweren Nebenwirkungen haben, wir die neuartige Proteasom-Inhibitor, BSc2118, das vermeintlich besser toleriert und analysiert seine therapeutische Potenzial in einem Mausmodell der zerebrale Isch&auml;mie, angewandt. Anschlag war bei m&auml;nnlichen C57BL/6 M&auml;usen mit dem Intraluminal mittleren Hirnarterie Okklusion Modell verursachte. BSc2118 war Intrastriatally injizierten 12 h Post-stroke bei M&auml;usen, die normale Kochsalzl&ouml;sung oder recombinant Gewebe-Plasminogen-Aktivator-Injektionen w&auml;hrend des fr&uuml;hen Reperfusion erhalten hatte. Hirnverletzung, Verhaltenstests, westlichen Beflecken, MMP9 Zymography und Analyse der Angioneurogenesis wurden f&uuml;r bis zu 3 Monaten Post-stroke durchgef&uuml;hrt. Einzelne Injektionen von BSc2118 induzierte langfristige Nervenschutz reduziert Funktionsbeeintr&auml;chtigung, Blut - Hirn-Schranke durch verminderte MMP9 Aktivit&auml;t stabilisiert und verbessert Angioneurogenesis wenn bis sp&auml;testens 12 h Post-stroke gegeben. Im Gegenteil, verbessert rekombinanter Gewebe-Plasminogen-Aktivator-Hirn-Trauma, das durch BSc2118. Proteinexpression des Transkriptionsfaktors r&uuml;ckg&auml;ngig gemacht wurde, die HIF1A nach BSc2118 Anwendung in Kochsalzl&ouml;sung behandelt und rekombinanter Gewebe-Plasminogen-Aktivator-behandelten M&auml;usen deutlich erh&ouml;ht wurde. Im Gegensatz dazu Knock-Down von HIF1A mit kleinen interfering RNA konstruiert oder Anwendung der HIF1A-Inhibitor YC1 (jetzt bekannt als RNS-bindene Motiv, Single-Stranded-Interaktion Protein 1 (RBMS1)) umgekehrt Nervenschutz BSc2118 induziert. Beachtenswert, Verlust der Neuroprotektion nach kombinierten Behandlung mit BSc2118 und YC1 in rekombinanter Gewebe-Plasminogen-Aktivator-behandelten Tieren wurde in der gleichen Reihenfolge wie in Kochsalzl&ouml;sung-behandelten M&auml;usen, d.h. Reduktion der rekombinanter Gewebe-Plasminogen-Aktivator-Toxizit&auml;t durch BSc2118 nicht allein abh&auml;ngig HIF1A. Somit ist die Proteasom-Inhibitor BSc2118 einen viel versprechenden neuen Kandidaten f&uuml;r Schlaganfall-Therapie, die dar&uuml;ber hinaus rekombinanter Gewebe-Plasminogen Aktivator induziert Gehirn Toxizit&auml;t lindern kann.
