Somatischer Zellen wird die Translokation von PKCs durch Rezeptor f&uuml;r aktivierte C-Kinase (RACK); erleichtert. jedoch seine Beteiligung an Ei Aktivierung noch schwer fassbar ist. Wir folgten das Translokation-Muster von konventioneller und neuartiger PKCs (cPKCs und nPKCs, beziehungsweise) auf Ei-Aktivierung. Konfokale Mikroskopie angegeben, den Ausdruck und die Lokalisierung von RACK1, einen spezifischen Rezeptor-Protein f&uuml;r cPKCs. Aktivierung des MII-Eier, f&uuml;hrte zur Translokation, die Ei-Kortex PKC&alpha;, &beta;II und &delta; und die Co-translocation Rack 1, mit PKC&alpha; und PKC&beta;II. Co-Immunopr&auml;zipitation zeigte der Verband der PKC und Actin, beide bekannt in kortikalen Granulat Exozytose (CGE) mit RACK1, einbezogen werden. Ei Aktivierung ergab eine erh&ouml;hte RACK1 zusammen mit einem verringerten Ma&szlig; an PKC&beta;II. Auf der Grundlage dieser Ergebnisse empfehlen wir, dass nach der Aktivierung Ei RACK1 Shuttles cPKCs, die Ei-Rinde, aktiviert und erleichtert so CGE.

Rezeptor f&uuml;r C-Kinase (RACK) und Proteinkinase C (PKC) in ei Aktivierung aktiviert.

In somatic cells, translocation of PKCs is facilitated by receptor for activated C kinase (RACK); however its involvement in egg activation is still elusive. We have followed the translocation pattern of conventional and novel PKCs (cPKCs and nPKCs, respectively) upon egg activation. Confocal microscopy indicated the expression and localization of RACK1, a specific receptor protein for cPKCs. Activation of MII eggs, led to translocation to the egg cortex of PKCÎ±, Î²II and Î´ and the co-translocation of RACK1, with both PKCÎ± and PKCÎ²II. The association of PKC and actin, both known to be involved in cortical granules exocytosis (CGE) with RACK1, was demonstrated by co-immunoprecipitation. Egg activation resulted in an increased RACK1 level along with a decreased level of PKCÎ²II. Based on these results, we suggest that upon egg activation, RACK1 shuttles activated cPKCs to the egg cortex, thus facilitating CGE.

Receptor for activated C kinase (RACK) and protein kinase C (PKC) in egg activation.

Mammalian glycogen synthase kinase-3 (GSK-3), a critical regulator in neuronal signaling, cognition, and behavior, exists as two isozymes GSK-3Î± and GSK-3Î². Their distinct biological functions remains largely unknown. Here, we examined the evolutionary significance of each of these isozymes. Surprisingly, we found that unlike other vertebrates that harbor both GSK-3 genes, the GSK-3Î± gene is missing in birds. GSK-3-mediated tau phosphorylation was significantly lower in adult bird brains than in mouse brains, a phenomenon that was reproduced in GSK-3Î± knockout mouse brains. Tau phosphorylation was detected in brains from bird embryos suggesting that GSK-3 isozymes play distinct roles in tau phosphorylation during development. Birds are natural GSK-3Î± knockout organisms and may serve as a novel model to study the distinct functions of GSK-3 isozymes.

S&auml;ugetier-Glykogen-Synthase Kinase-3 (GSK-3), einen kritischen Regulator in neuronale Signale, Kognition und Verhalten, vorhanden als zwei Isoenzyme GSK-3&alpha; und GSK-3&beta;. Ihre eindeutigen biologischen Funktionen bleibt weitgehend unbekannt. Hier haben wir die evolution&auml;re Bedeutung der einzelnen dieser Isoenzyme gepr&uuml;ft. &Uuml;berraschenderweise fanden wir, dass im Gegensatz zu anderen Wirbeltieren, die beide GSK-3-Gene zu beherbergen, das GSK-3&alpha;-gen bei V&ouml;geln fehlt. GSK-3-vermittelter Tau Phosphorylierung war Erwachsenen Vogel Gehirne erheblich niedriger als in Maus-Gehirn, Gehirn, ein Ph&auml;nomen, das in GSK-3&alpha;-Knockout-Maus reproduziert wurde. Tau Phosphorylierung wurde festgestellt, in den Gehirnen von Vogel-Embryos, die darauf hindeutet, dass GSK-3 Isoenzyme unterschiedliche Rollen in Tau Phosphorylierung w&auml;hrend der Entwicklung spielen. V&ouml;gel sind nat&uuml;rliche Organismen, GSK-3&alpha; k.o. und dienen als neuartige Modell die unterschiedlichen Funktionen der GSK-3 Isoenzyme zu studieren.
