During natural HIV infection, an array of host receptors are thought to influence virus attachment and the kinetics of infection. In this study, to probe the interactions of HIV envelope (Env) with various receptors, we assessed the inhibitory properties of various anti-Env monoclonal antibodies (mAbs) in binding assays. To assist in detecting Env in attachment assays, we generated Fc fusions of full-length wild-type gp120 and several variable loop-deleted gp120s. Through investigation of the inhibition of Env binding to cell lines expressing CD4, CCR5, DC-SIGN, syndecans or combinations thereof, we found that the broadly neutralizing mAb, 2G12, directed to a unique carbohydrate epitope of gp120, inhibited Env-CCR5 binding, partially inhibited Env-DC-SIGN binding, but had no effect on Env-syndecan association. Furthermore, 2G12 inhibited Env attachment to primary monocyte-derived dendritic cells, that expressed CD4 and CCR5 primary HIV receptors, as well as DC-SIGN, and suggested that the dual activities of 2G12 could be valuable in vivo for inhibiting initial virus dissemination and propagation.

W&auml;hrend nat&uuml;rliche HIV-Infektion ein Array von Host-Rezeptoren werden gedacht, um Virus-Anlage und die Kinetik der Infektion zu beeinflussen. Natural regulatory T cells (Tregs) are present in high frequencies among tumor-infiltrating lymphocytes and in draining lymph nodes, supposedly facilitating tumor development. To investigate their role in controlling local immune responses, we analyzed intratumoral T cell accumulation and function in the presence or absence of Tregs. Tumors that grew in normal BALB/c mice injected with the 4T1 tumor cell line were highly infiltrated by Tregs, CD4 and CD8 cells, all having unique characteristics. Most infiltrating Tregs expressed low levels of CD25Rs and Foxp3. They did not proliferate even in the presence of IL-2 but maintained a strong suppressor activity. CD4 T cells were profoundly anergic and CD8 T cell proliferation and cytotoxicity were severely impaired. Depletion of Tregs modified the characteristics of tumor infiltrates. Tumors were initially invaded by activated CD4(+)CD25(-) T cells, which produced IL-2 and IFN-gamma. This was followed by the recruitment of highly cytotoxic CD8(+) T cells at tumor sites leading to tumor rejection. The beneficial effect of Treg depletion in tumor regression was abrogated when CD4 helper cells were also depleted. These findings indicate that the massive infiltration of tumors by Tregs prevents the development of a successful helper response. The Tregs in our model prevent Th cell activation and subsequent development of efficient CD8 T cell activity required for the control of tumor growth.

Nat&uuml;rliche regulatorische T-Zellen (Tregs) sind vorhanden in hohen Frequenzen zwischen den Tumor-infiltrieren Lymphozyten und Entw&auml;sserung Lymphknoten, Tumor-Entwicklung angeblich zu f&ouml;rdern. Um ihre Rolle bei der Kontrolle der lokalen Immunantworten zu untersuchen, analysiert wir intratumorale T-Zell-Akkumulation und Funktion im Vorhandensein oder Nichtvorhandensein von Tregs. Tumoren, die in normalen BALB/c M&auml;usen injiziert mit der 4T1 Tumor-Zelllinie wuchs stark infiltriert wurden Tregs, CD4 und CD8-Zellen, alle, die einzigartige Eigenschaften. Die meisten infiltrierendes Tregs ausgedr&uuml;ckt niedrige CD25Rs und Foxp3. Sie nicht vermehren sich auch in Gegenwart von Il aber eine starke Tumorsuppressor-Aktivit&auml;t beibehalten. CD4 T-Zellen waren zutiefst Anergic und CD8-T-Zellproliferation und Zytotoxizit&auml;t stark eingeschr&auml;nkt. Ersch&ouml;pfung der Tregs ver&auml;ndert die Eigenschaften der Tumor infiltriert. Tumoren wurden zun&auml;chst aktivierte CD4(+)CD25(-) T-Zellen, drangen die Il und IFN-Gamma produziert. Danach folgte die Einstellung von hoch zytotoxischen CD8(+) T-Zellen bei Tumor zu Tumor Ablehnung f&uuml;hrt. Die wohltuende Wirkung der Treg Ersch&ouml;pfung bei der Tumor-Regression wurde aufgehoben, wenn CD4-Helferzellen auch ersch&ouml;pft waren. Diese Befunde zeigen, dass das massive Eindringen von Tumoren von Tregs verhindert, die Entwicklung einer erfolgreichen Helfer-Antwort dass. Tregs in unserem Modell Th-ZELLAKTIVIERUNG und nachfolgende Entwicklung von effizienten CD8-T-Zellaktivit&auml;t ben&ouml;tigt, zur Steuerung von Tumorwachstum verhindern.
